Bio burden Membrane Filtration Method

The bioburden membrane filtration method is a microbiology test. the packaging material is a Laminate tube and aluminum tube. it is suitable for finishing products, bacterial growth, or fungal growth.

Select twenty samples at random from the sample lot

Accessories required for testing

  • 20 sample of tubes
  • 1% sterile peptone
  • Sterile soybean casein digest agar plate
  • Sterile membrane filter
  • Sabourauds chloramphenicol agar

Procedure

  • Aseptically place 4 ml of sterile 0.1% peptone in each of the 10 samples of the aluminium tube from the open end, fold the end, and shake the tube thoroughly.
  • Collect the rinsing solution from each tube and filter the pooled solution through a sterile membrane filter.
  • Place the membrane filter aseptically on the surface of a sterile pre-incubated casein digest agar plate
  • Incubated the above plate at 300c – 350c for day in inverted position.
  • Count the colonies observed on the membrane filter and report the total bacterial count as ‘X ‘ cfu/ 10 units.
  • Repeat the above steps for the fungal count by using Sabourauds chloramphenicol agar and incubate the plate 200c – 250c for 5 days
  • Count the colonies observed on the membrane filter and report the total fungal counts as ‘y’ cfu/10 units
